ABSTRACT
Background: Klebsiella pneumoniae is a bacterium that is known to significantly cause various infections in humans. This study focused on investigating the morphology, the results of biochemical tests, and total plate number of lyophilizated Klebsiella pneumoniae bacteria with horse serum as a lipoprotectant before and after preserved at room temperature for 2 months.
Objective: The purpose of this study was to determine the effectiveness of the lyophilization method with horse serum lipoprotectants by preserving it at room temperature for two months to maintain morphology, biochemical test results and bacterial viability.
Methods: This type of research is interventional research with repeated observation research design. The subject used was the bacterium Klebsiella pneumonia ATCC 33495. Lipoprotectant was added in the form of horse serum before the lyophilization process. After the lyophilization process is complete, before being stored for two months, morphological observations, biochemical tests and total plate number calculations are carried out first. After the two-month storage period is complete, observations are carried out again as before. The results of the data from the two observations were then compared to see how the condition of the bacteria was before and after being stored for two months.
Results: The results of research and preserving for two months showed that the preserved Klebsiella pneumoniae bacteria could maintain its morphological characteristics and biochemical tests and showed sustainable metabolic activity. However, the viability results obtained were 0.093% with a decrease in the total plate number yield of 99.95%. Nonetheless, this study still demonstrates the potential of lyophilization as a storage method for Klebsiella pneumoniae bacteria, which could have significant implications in various fields including diagnostics and clinical research.
Conclusion: The results showed that most of the bacteria in the samples were not able to maintain their life during the preservation process. Therefore, the lyophilized preserve method with horse serum lipoprotectants and preserved for two months is less effective to do.
