Background : Microscopic examination is the standard method for diagnosing malaria. In this examination, blood samples are stained with Giemsa dye to see the morphology of blood cells, parasite cells, and the growth phase of Plasmodium. However, some areas are difficult to obtain the Giemsa dye buffer solution that is usually used. Therefore, this study aimed to evaluate the use of 0.9% NaCl solution as a diluent and pH 7.2 buffer solution for Giemsa staining in Plasmodium vivax malaria blood tests.
Objective : To determine whether 0.9% NaCl solution can be used as a substitute for phosphate buffer pH 7.2 in the Giemsa staining of malaria smears.
Methods : The research methodology employed is quasi-experimental. The samples utilised are malaria blood preparations stained with Giemsa, which have been diluted with a 0.9% NaCl solution. The data obtained from microscopic observations were subjected to descriptive analysis using the inter-rater reliability method with the Cohen’s Kappa coefficient.
Results : From these results, it can be seen that the reliability of the results obtained for the assessment of parasite nuclei in thin smears was 0.148, while the reliability of the results obtained for the assessment of parasite cytoplasm in thin smears was 0.406. Furthermore, the reliability of the results obtained for the assessment of parasite nuclei in thick smears was 0.631, while the reliability of the results obtained for the assessment of parasite cytoplasm in thick smears was 0.460.
Conclusion : NaCl 0.9% solution can be used as a diluent in Giemsa staining of malaria blood preparations.
